About 17,000 runners are expected to take part in events on the first weekend of May.
A new city-centre finish has been unveiled for the AJ Bell Great Birmingham Run, which organisers say is one of the UK's largest mass participation running events.
Thousands of runners are expected to take part across the weekend of Saturday 2 and Sunday 3 May.
On Sunday, the half marathon and 10k routes will once again "showcase Birmingham at its best", starting in Centenary Square and passing major landmarks including the Jewellery Quarter, Grand Central and St Martin's Church, organisers said.
This year, runners will cross the line at a new "festival-style" finish village at Aston University, which organisers say marks an exciting update to the event route.
More than 17,000 are expected to take part across the weekend with the junior and mini Great Birmingham Run returning to Alexander Stadium on the Saturday.
Paul Foster, chief executive of the Great Run Company, said: "This year's event is shaping up to be one of our most exciting yet.
"The new finish along Jennens Road and into Aston University allows us to create a real celebration for runners, their supporters and our charity partners."
Aston University's vice chancellor Aleks Subic added: "It is fitting that the university was chosen because 2026 marks our 60th anniversary, which we will be marking with a year of celebrations such as the Aston Made campaign, which reflects the impact Aston University has had on Birmingham and the rest of the world."
